== History == Anti-U1 RNP antibodies were first described in the early 1970s during studies of patients with mixed connective tissue disease. In 1972, rheumatologist Dr, Gordon Sharp and colleagues identified antibodies directed against extractable nuclear antigens (ENAs) in patients who had symptoms of several autoimmune connective tissue diseases. Their work helped establish mixed connective tissue disease (MCTD) as a distinct clinical syndrome and made anti-U1 RNP antibodies an important laboratory discovery in rheumatology. The discovery of anti-U1 RNP antibodies also improved the understanding of autoimmune connective tissue disease. Before these antibodies were identified, many patients with overlapping symptoms were difficult to classify because their conditions shared characteristics of several different autoimmune disorders. Identifying anti-U1 RNP antibodies gave healthcare physicians another way to recognize patterns of disease and support the diagnosis of mixed connective tissue disease. Although the antibodies are not exclusive to MCTD, they remain one of the most characteristic scientific findings associated with the condition. Sanger sequencing A method of DNA sequencing based on the in vitro replication of a DNA template sequence, during which fluorochrome-labeled, chain-terminating dideoxynucleotides are randomly incorporated in the elongating strand; the resulting fragments are then sorted by size with electrophoresis, and the particular fluorochrome terminating each of the size-sorted fragments is detected by laser chromatography, thus revealing the sequence of the original DNA template through the order of the fluorochrome labels as one reads from small-sized fragments to large-sized fragments. Though Sanger sequencing has been replaced in some contexts by next-generation methods, it remains widely used for its ability to produce relatively long sequence reads (500+ nucleotides) and its very low error rate.

By the end of the 19th century, Cossack communities enjoyed a privileged tax-free status in the Russian Empire, although they had a 20-year military service commitment (reduced to 18 years from 1909). They were on active duty for five years, but could fulfill their remaining obligation with the reserves. At the beginning of the 20th century, the Russian Cossacks numbered 4.5 million. They were organized as independent regional hosts, each comprising a number of regiments. The need for the government to call up Cossack men to serve either with the Army or a mounted police force caused many social and economic problems, which compounded by the growing impoverishment the communities of the Hosts. Treated as a separate and elite community by the Tsar, the Cossacks rewarded his government with strong loyalty. His administration frequently used Cossack units to suppress domestic disorder, especially during the Russian Revolution of 1905. The Imperial Government depended heavily on the perceived reliability of the Cossacks. By the early 20th century, their decentralized communities and semi-feudal military service were coming to be seen as obsolete. The Russian Army Command, which had worked to professionalize its forces, considered the Cossacks less well disciplined, trained, and mounted than the hussars, dragoons, and lancers of the regular cavalry. The Cossack qualities of initiative and rough-riding skills were not always fully appreciated. As a result, Cossack units were frequently broken up into small detachments for use as scouts, messengers, or picturesque escorts.
