Pharmacokinetic accounts emphasize rapid breakdown. After intravenous dosing the intact peptide disappears from blood within minutes, and nasal delivery produces low but measurable concentrations. Metabolites rather than the parent molecule may account for part of the observed activity, although the relative contribution is unresolved. Dosing in the literature varies widely and no optimal schedule has been agreed. These gaps are regularly cited as a reason the findings have not produced broad clinical adoption beyond the original research setting.
Proposed mechanisms center on neurotrophic signaling rather than on classical melanocortin receptor activation. Rodent experiments have reported shifts in the expression of brain-derived neurotrophic factor and nerve growth factor after administration, together with changes in the associated receptor systems. Several authors argue that the peptide acts largely through its degradation products and their interaction with peptidergic pathways, but this remains a hypothesis rather than a settled finding. No single molecular target has been identified in a way that the field broadly accepts.
Claims about enhanced focus, memory, or mood in healthy people rest mostly on anecdotal reports and community discussion rather than on controlled data. It remains unclear whether any cognitive benefit observed in patients recovering from brain injury would extend to uninjured users. Dose-response relationships, long-term safety, and interactions with other drugs are not well characterized in the peer-reviewed literature. Questions about optimal route of administration and treatment duration likewise remain open.
Semax binds to melanocortin receptors and is thought to influence neuronal survival and plasticity rather than to act through the adrenal axis. Laboratory work has shown increased expression of brain-derived neurotrophic factor and nerve growth factor in treated tissue. Changes in c-Fos, a marker of neuronal activation, have also been reported. Because the peptide is rapidly degraded by peptidases, its effects are generally attributed to downstream signaling cascades rather than to sustained receptor occupancy.
Published studies are dominated by animal models of stroke, ischemia, and cognitive impairment, with a smaller number of human trials conducted in Russia. Many of the human reports are small, single-center, and published in Russian-language journals, which limits independent scrutiny. Outcome measures vary between studies and often rely on clinician-rated scales rather than objective biomarkers. Systematic reviews have noted the methodological weaknesses and called for larger, preregistered trials before firm conclusions can be drawn.
| Property | Value | Notes |
|---|---|---|
| Primary research models | Rodent studies | Largest share of published data |
| Reported markers | BDNF and NGF expression | Measured mainly in animal tissue |
| Common administration | Intranasal | Matches the registered formulation |
| Blood residence time | Minutes | Rapid enzymatic degradation |
| Evidence quality | Small trials, limited replication | Noted repeatedly in reviews |
Circulation time for the peptide is short because peptidases cleave it readily. The Pro-Gly-Pro tail is thought to slow breakdown compared with the bare ACTH fragment, but the gain appears modest. Absorption after intranasal dosing is limited, and only a fraction of a dose is expected to reach the central nervous system. Laboratory concentrations therefore sit well above levels achieved systemically, a gap that complicates translation from bench findings to clinical claims.
The mechanisms attributed to semax are inferred from animal and cell studies rather than traced to one confirmed target. The most frequently cited pathway involves increased expression of brain-derived neurotrophic factor and nerve growth factor in hippocampal and cortical tissue. Some work points to engagement of melanocortin receptors, particularly MC4, which the parent ACTH fragment can activate. Effects on dopaminergic and serotonergic signalling have also been reported. No single account explains all observed results, and the relative weight of each pathway remains unsettled.
Published studies examine a fairly narrow set of endpoints. Rodent experiments commonly measure maze learning, infarct volume after induced ischemia, and tissue levels of neurotrophic factors. Clinical reports from Russian centres describe attention, memory and recovery scores in patients after stroke or transient ischemic attack. Most of those human studies are small and few have been repeated by independent groups. Outcome measures differ between studies, which limits direct comparison.
== Metabolism == Vicine is an inactive compound in the body. When vicine enters the body through food, it is hydrolysed by intestinal microflora to the highly reactive free radical generating aglycone divicine. Upon hydrolysis, the glucose part of the molecule is split off and that results in the reduced divicine. Divicine is then taken up in the blood through the intestinal epithelium.
It is rare to find less than five interacting parallel strands in a motif, suggesting that a smaller number of strands may be unstable, however it is also fundamentally more difficult for parallel β-sheets to form because strands with N and C termini aligned necessarily must be very distant in sequence . There is also evidence that parallel β-sheet may be more stable since small amyloidogenic sequences appear to generally aggregate into β-sheet fibrils composed of primarily parallel β-sheet strands, where one would expect anti-parallel fibrils if anti-parallel were more stable. In parallel β-sheet structure, if two atoms Cαi and Cαj are adjacent in two hydrogen-bonded β-strands, then they do not hydrogen bond to each other; rather, one residue forms hydrogen bonds to the residues that flank the other (but not vice versa). For example, residue i may form hydrogen bonds to residues j − 1 and j + 1; this is known as a wide pair of hydrogen bonds. By contrast, residue j may hydrogen-bond to different residues altogether, or to none at all. The hydrogen bond arrangement in parallel beta sheet resembles that in an amide ring motif with 11 atoms. Finally, an individual strand may exhibit a mixed bonding pattern, with a parallel strand on one side and an antiparallel strand on the other.
The treatment of slaves in the United States varied widely depending on conditions, times, and places. The power relationships of slavery corrupted many whites who had authority over slaves, with children showing their own cruelty. Masters and overseers resorted to physical punishments to impose their wills. Slaves were punished by whipping, shackling, hanging, beating, burning, mutilation, branding and imprisonment. Punishment was most often meted out in response to disobedience or perceived infractions, but sometimes abuse was carried out to re-assert the dominance of the master or overseer of the slave. Treatment was usually harsher on large plantations, which were often managed by overseers and owned by absentee slaveholders. William Wells Brown, who escaped to freedom, reported that on one plantation, slave men were required to pick 80 pounds (36 kg) of cotton per day, while women were required to pick 70 pounds (32 kg) per day; if any slave failed in their quota, they were subject to whip lashes for each pound they were short. The whipping post stood next to the cotton scales. A New York man who attended a slave auction in the mid-19th century reported that at least three-quarters of the male slaves he saw at sale had scars on their backs from whipping. By contrast, small slave-owning families had closer relationships between the owners and slaves; this sometimes resulted in a more humane environment but was not a given.
=== Anabaptist === Mennonite Mutual Aid Association - Founded in July 1945 as Mennonite Mutual Aid. In its first ten years, it expended loans to Civilian Public Service workers following their service in World War II. In the following 15 years, different insurance programs were established, including automobile insurance, hospital and burial benefits, etc. Became a fraternal benefit society in January 1966 as the Mennonite Mutual Aid Association. Merged with other Mennonite financial union groups to become Everence in 2010. Originally open to Mennonites 16 and up, the association's denominational scope was enlarged to include other Anabaptist denominations in 1984. Today Everence, while remaining the stewardship agency of the Mennonite Church USA, offers its "products and services ... to everyone interested in practicing stewardship that aligns with our founding values." In 1979 locals were called "Branches" and were usually affiliated with congregations of the Mennonite General Conference. Each branch must meet at least 12 times a year. The highest authority was the "Biennial Conference". There were also district conferences. February 1979 membership 4,984 in 12 states. Sponsored seminars on church leadership, family life training, estate planning, financial counseling, youth leadership, and family communications. Programs include helping local families with emergency needs, aiding local church projects;
=== Internal nasal anatomy === In the midline of the nose, the septum is a composite (osseo-cartilaginous) structure that divides the nose into two similar halves. The lateral nasal wall and the paranasal sinuses, the superior concha, the middle concha, and the inferior concha, form the corresponding passages, the superior meatus, the middle meatus, and the inferior meatus, on the lateral nasal wall. The superior meatus is the drainage area for the posterior ethmoid bone cells and the sphenoid sinus; the middle meatus provides drainage for the anterior ethmoid sinuses and for the maxillary and frontal sinuses; and the inferior meatus provides drainage for the nasolacrimal duct. The internal nasal valve comprises the area bounded by the upper lateral-cartilage, the septum, the nasal floor, and the anterior head of the inferior turbinate. In the narrow (leptorrhine) nose, this is the narrowest portion of the nasal airway. Generally, this area requires an angle greater than 15 degrees for unobstructed breathing; for the correction of such narrowness, the width of the nasal valve can be increased with spreader grafts and flaring sutures.

=== NAD+ pptimization === 1-MNA is an inhibitor of nicotinamide N-methyltransferase (NNMT). By inhibiting NNMT activity, it regulates NAD+ biosynthesis via the nicotinamide salvage pathway, the primary route for NAD+ synthesis in mammals. By participating in this pathway, 1-MNA optimizes NAD+ levels.
Somalia contains a variety of mammals due to its geographical and climatic diversity. Wildlife still occurring includes cheetah, lion, reticulated giraffe, baboon, serval, elephant, bushpig, gazelle, ibex, kudu, dik-dik, oribi, Somali wild ass, reedbuck and Grévy's zebra, elephant shrew, rock hyrax, golden mole and antelope. It also has a large population of the dromedary camel. Somalia is home to around 727 species of birds. Of these, eight are endemic, one has been introduced by humans, and one is rare or accidental. Fourteen species are globally threatened. Birds species found exclusively in the country include the Somali pigeon; Alaemon hamertoni (Alaudidae), Lesser Hoopoe-Lark; Heteromirafra archeri (Alaudidae), Archer's Lark; Mirafra ashi, Ash's Bushlark; Mirafra somalica (Alaudidae), Somali Bushlark; Spizocorys obbiensis (Alaudidae), Obbia Lark; Carduelis johannis (Fringillidae); and Warsangli Linnet. Somalia's territorial waters are prime fishing grounds for highly migratory marine species, such as tuna. A narrow but productive continental shelf contains several demersal fish and crustacean species. Fish species found exclusively in the nation include Cirrhitichthys randalli (Cirrhitidae), Symphurus fuscus (Cynoglossidae), Parapercis simulata OC (Pinguipedidae), Cociella somaliensis OC (Platycephalidae), and Pseudochromis melanotus (Pseudochromidae). There are roughly 235 species of reptiles. Of these, almost half live in the northern areas.

== History == Cell display systems were first used in 1985, when peptides were genetically fused with proteins displayed on the M13 bacteriophage. Bacteriophage display is a commonly used cell display system, although it carries limitations in the size of proteins that can be displayed. Bacterial display was then introduced in 1986, allowing the surface display of larger proteins. Bacterial display systems were first introduced by Freudl et al. and Charbit et al. in 1986, when they used bacterial surface proteins OmpA and LamB to display peptides. Freudl et al. fused peptides with linkers with the ompA gene, causing the peptides to be expressed in the OmpA proteins. They showed that the proteins were now subject to cleavage by proteinase K. The non-OmpA peptides inserted were therefore a target of proteinase K. Insertion of the foreign peptides did not affect bacterial cell growth. Charbit et al. firstly defined the areas of the LamB protein that were "permissive" for foreign petide insertion (ie that did not lead to a complete loss of functionality of the protein). Then, they explored the versatility of the permissive sites (size limit, nature of the epitope,...) that were all located in surface-exposed loops of the trimeric outer membrane porin, aiming at developing multivalent live bacterial vaccines. This was the first evidence of using bacterial surface display techniques to express proteins on the surface of cells, without altering the function of the cell.
== Alcohol and beta blockers == Alcohol, more precisely ethanol, is a colorless flammable liquid that is the intoxicating agent in fermented and distilled liquors. Beta blockers are any of a group of drugs that decrease the rate and force of heart contractions and lower high blood pressure. Beta blockers help athletes in sports such as rifle shooting or archery where nervousness can harm performance, and they are only banned in these sports. Examples beta blockers are atenolol, metoprolol, nadolol, pindolol, propranolol, and timolol.

The parent compound of this group of substances, pyridine, was obtained by Thomas Anderson around 1849 during the dry distillation of bones. Anderson also isolated the picolines (methylpyridines) from coal tar and bone oil. The structure of the pyridine ring was elucidated around 1870 by Wilhelm Körner and James Dewar.
=== Becky Conner === Rebecca "Becky" Conner (later Healy) is played by Lecy Goranson (Sarah Chalke in seasons 6–9). Born in 1975, Becky is the eldest of Roseanne and Dan's children. Becky is introduced to the series as a pre-teen, whose primary interests are centered around makeup, fashion, and boys. While she can be self-centered and occasionally acts spoiled, she actually is the least problematic of the Conner children, maintaining high grades and rarely causing issues for her parents beyond an occasional argument. However, as Becky matures, she grows moody and rebellious, resulting in her and a friend getting drunk on Roseanne and Dan's alcohol while alone in the Conner house. She has several clean-cut boyfriends, but soon prefers dating edgier, punkish guys that Roseanne and Dan disapprove of. They especially dislike her latest boyfriend (and future husband), Mark Healy, particularly after the young couple become sexually active. Becky's ultimate rebellion comes when she is seventeen and drops out of high school to elope with Mark and move to Minneapolis. In the year before her elopement, Becky's life was stressful. In addition to schoolwork and working as a cashier to save for a car, she was responsible for maintaining the household and tending her younger siblings while her parents worked long hours. Her breaking point came when she learnt her parents had used her college fund to pay household bills and, finally, when the family's bike shop failed, forcing her boyfriend Mark, who worked there, to move to Minneapolis for a new job.
=== Non-competitive === In non-competitive inhibition the binding of the inhibitor to the enzyme reduces its activity but does not affect the binding of substrate. This type of inhibitor binds with equal affinity to the free enzyme as to the enzyme-substrate complex. It can be thought of as having the ability of competitive and uncompetitive inhibitors, but with no preference to either type. As a result, the extent of inhibition depends only on the concentration of the inhibitor. Vmax will decrease due to the inability for the reaction to proceed as efficiently, but Km will remain the same as the actual binding of the substrate, by definition, will still function properly.
== Mental health == The high rate of smoking tobacco by people with mental health problems is a major factor in their decreased life expectancy, which is about 25 years shorter than that of the general population.
=== 19 May === The Ukrainian military said it had repelled another Russian air attack in the morning, destroying 19 drones and three Kalibr missiles out of a total of 28 launched. An elderly woman was seriously injured during airstrikes on Kryvyi Rih. The Pentagon announced that it had made an accounting error in relation to the aid supplied to Ukraine. The error was made when the Pentagon overestimated to Congress, by at least $3 billion, the cost of weapons sent to Ukraine. This error meant that these funds were effectively a surplus that could be spent on more weapons for Ukraine. President Biden said he would support a joint effort with allies to train Ukrainian pilots on fourth generation aircraft, including F-16s fighter jets. Shortly afterwards, Denmark said it would provide direct help in training. No date was announced for such training to commence. Once it does it could take six to nine months to properly train Ukrainian pilots and crews to use the F-16. A US official told CNN that the United States was prepared to help train Ukrainian pilots on the F-16. The training would take place in Europe in the "coming weeks", along with European allies, while a decision on the number of aircraft would be made at a later date. During the 49th G7 summit in Hiroshima, Japan, UK Prime Minister Rishi Sunak announced that the government was introducing new sanctions against Russian exports, particularly on diamonds, nickel, copper and aluminum.

The main proposal is modulation of neurotrophic factors such as brain-derived neurotrophic factor, supported largely by animal experiments. Receptor-level targets have not been firmly established. Most reviews describe the mechanism as only partially characterized.
The compound is registered as a medicine in Russia, so clinical work has concentrated there. Trials elsewhere are few and generally small. This geographic concentration is a recognized limitation in evidence reviews.
Some fraction may reach the central nervous system through olfactory pathways, and this is often cited as the rationale for the nasal route. The size of that fraction in humans is not well quantified. Blood concentrations after nasal dosing are low, which complicates measurement.
It is associated with the melanocortin receptor family, particularly subtypes found in the central nervous system. This interaction is separate from the adrenal pathway activated by full-length ACTH.
