Semax raises a handful of sensible questions. This page answers them in order, starting with the fundamentals and moving to applications.
Reviewed 2025-09-16. Anything still debated is marked as such rather than presented as settled.
The C-terminal Pro-Gly-Pro extension is not incidental. Proline-rich tails are known to resist several common peptidases, and the published literature attributes the longer half-life of Semax, relative to unmodified ACTH fragments, to this feature. The modification also removes the melanocyte-stimulating and corticosteroidogenic activity that characterizes longer ACTH-derived sequences. Because the molecule is small and hydrophilic, it is typically formulated as an aqueous solution for intranasal or parenteral delivery. Acetylation or amidation at the termini appears in closely related research peptides and shifts the mass by a fixed increment.
Reported pharmacological work centers on neurotrophic signaling, including changes in BDNF and NGF expression in hippocampal tissue in animal models. Human data come largely from studies conducted in Russia, and how well those results generalize to other populations remains an open question. Regulatory status differs sharply by jurisdiction: Semax is a registered prescription medicine in Russia, while it holds no approved marketing status in the United States or the European Union. Outside such jurisdictions it is generally handled as a research chemical, which affects both documentation and quality expectations.
Terminology around the compound varies by source. It appears in catalogues and papers as Semax, as the heptapeptide ACTH(4-7)-Pro-Gly-Pro, and under various alphanumeric laboratory codes used by individual suppliers. These names refer to the same sequence but may imply different salt forms, purity grades, or counter-ions. Peptide databases usually list the free base mass, while product descriptions sometimes report acetate or trifluoroacetate salts with a different formula weight. Because naming conventions for research peptides are not standardised across vendors, checking the declared sequence and measured mass is more reliable than relying on a trade name alone.
Semax is a synthetic heptapeptide with the sequence Met-Glu-His-Phe-Pro-Gly-Pro. Its design combines the ACTH(4-7) core fragment with a C-terminal Pro-Gly-Pro extension, a modification intended to improve stability and prolong activity. The molecule is hydrophilic, carries no lipid chains or glycosylation, and has a theoretical mass just over 810 daltons in its free form. All seven residues are proteinogenic amino acids, so no non-natural building blocks appear in the backbone. A free N-terminal methionine and C-terminal proline define the unmodified parent peptide.
| Property | Value | Notes |
|---|---|---|
| Molecular weight | ~813.9 Da (free heptapeptide) | N-acetylated form is about 42 Da heavier |
| Residue count | 7 (heptapeptide) | Four residues from ACTH(4-10), three added |
| Appearance | White to off-white powder | Usually supplied lyophilized |
| Solubility class | Freely soluble in water | Also dissolves in DMSO and polar organic solvents |
| Related parent fragment | ACTH(4-10) | Parent motif is Met-Glu-His-Phe |
Semax is a synthetic heptapeptide with the sequence Met-Glu-His-Phe-Pro-Gly-Pro. It is described in the literature as an analogue of the ACTH(4–10) fragment, a short N-terminal portion of adrenocorticotropic hormone that retains some neurotropic activity without the full hormonal effects of the parent peptide. The molecule carries a methionine residue at the N-terminus and two proline residues near the C-terminus, features that shape both its interactions with receptor systems and its chemical stability. The free peptide corresponds to the formula C37H51N9O10S and a molecular mass near 813.9 Da.
The compound was developed in the 1980s at the Institute of Molecular Genetics in Moscow, where it emerged from research on short ACTH fragments and their effects on the central nervous system. Russian pharmaceutical listings describe it as a nootropic and neuroprotective agent, most often formulated as nasal drops. It is not a marketed medicine in the United States or the European Union, and no pharmacopoeial monograph covers it. Consequently, most published clinical experience with the substance originates from a small number of research centres, mainly in Russia and neighbouring countries.
Regulatory status varies sharply by country. Semax is registered for medical use in Russia, where it appears in formularies as a nasal solution, and it also holds registration in a small number of neighbouring states. It has no approval from the United States Food and Drug Administration or the European Medicines Agency, and it is not a scheduled controlled substance in most jurisdictions. Elsewhere it circulates mainly as laboratory material, so purity documentation comes from suppliers rather than from a national pharmacopoeia.
Semax is a synthetic peptide created in the Soviet Union during the early 1980s by researchers working in Moscow. It was built from the short adrenocorticotropic hormone fragment known as ACTH(4-10), and the chain was then extended with three additional amino acids. The resulting molecule was named semax and entered clinical use in Russia in 1994. It is generally described as a nootropic and neuroprotective agent rather than as a hormone analogue.
Laboratory work points toward modulation of neurotrophic signaling, particularly expression of brain-derived neurotrophic factor and nerve growth factor in hippocampal tissue. Studies also describe effects on monoamine turnover, inflammatory mediators, and oxidative markers. These observations come mainly from animal models and cultured cells, so the causal chain in humans is not firmly established. Whether the reported molecular changes translate into measurable clinical benefit is an open question. Reviews generally present the mechanism as plausible rather than demonstrated.
Clinical evidence consists largely of small trials with modest sample sizes, often without independent replication. Reported endpoints include cognitive scores, recovery after stroke, and visual function, but study designs vary widely and few trials meet contemporary reporting standards. Systematic reviewers have noted a high risk of bias in several of these reports. No large multicenter trial conducted outside Russia has been published. The compound is therefore best described as investigational in most jurisdictions, with its clinical role still unresolved.
=== Driving forces for moisture movement === Three main driving forces used in different version of diffusion models are moisture content, the partial pressure of water vapour, and the chemical potential of water. These are discussed here, including capillary action, which is a mechanism for free water transport in permeable softwoods. Total pressure difference is the driving force during wood vacuum drying.
l-DOPA, also known as l-3,4-dihydroxyphenylalanine and used medically as levodopa, is made and used as part of the normal biology of some plants and animals, including humans. Humans, as well as a portion of the other animals that utilize l-DOPA, make it via biosynthesis from the amino acid l-tyrosine. l-DOPA is the precursor to the neurotransmitters dopamine, norepinephrine (noradrenaline), and epinephrine (adrenaline), which are collectively known as catecholamines. Furthermore, l-DOPA itself mediates neurotrophic factor release by the brain and central nervous system. In some plant families (of the order Caryophyllales), l-DOPA is the central precursor of a biosynthetic pathway that produces a class of pigments called betalains. l-DOPA can be manufactured and in its pure form is sold as a drug with the INNTooltip International Nonproprietary Name levodopa. As a drug, it is used in the treatment of Parkinson's disease and dopamine-responsive dystonia, as well as restless leg syndrome. l-DOPA has a counterpart with opposite chirality, d-DOPA. As is true for many molecules, the human body produces only one of these isomers (the l-DOPA form). The enantiomeric purity of l-DOPA may be analyzed by determination of the optical rotation or by chiral thin-layer chromatography.
Afamelanotide (brand name Scenesse) has been approved for the treatment of erythropoietic protoporphyria in Europe and is also being investigated as a method of photoprotection in the treatment of polymorphous light eruption, actinic keratosis and squamous cell carcinoma (a form of skin cancer). An additional analogue called melanotan II causes enhanced libido and erections in most male test subjects and arousal with corresponding genital involvement in most female test subjects. Bremelanotide (formerly PT-141) which stemmed from melanotan II research is currently under development for its aphrodisiac effects. These effects are mediated by actions in the hypothalamus on neurons that express MC3 and MC4 receptors.
=== Structure-based === Structure-based drug design (or direct drug design) relies on knowledge of the three dimensional structure of the biological target obtained through methods such as x-ray crystallography or NMR spectroscopy. If an experimental structure of a target is not available, it may be possible to create a homology model of the target based on the experimental structure of a related protein. Using the structure of the biological target, candidate drugs that are predicted to bind with high affinity and selectivity to the target may be designed using interactive graphics and the intuition of a medicinal chemist. Alternatively, various automated computational procedures may be used to suggest new drug candidates. Current methods for structure-based drug design can be divided roughly into three main categories. The first method is identification of new ligands for a given receptor by searching large databases of 3D structures of small molecules to find those fitting the binding pocket of the receptor using fast approximate docking programs. This method is known as virtual screening. A second category is de novo design of new ligands. In this method, ligand molecules are built up within the constraints of the binding pocket by assembling small pieces in a stepwise manner. These pieces can be either individual atoms or molecular fragments. The key advantage of such a method is that novel structures, not contained in any database, can be suggested. A third method is the optimization of known ligands by evaluating proposed analogs within the binding cavity.
Nicholson at Birkbeck College, University of London and later at Imperial College London. In 1984, Nicholson showed 1H NMR spectroscopy could potentially be used to diagnose diabetes mellitus, and later pioneered the application of pattern recognition methods to NMR spectroscopic data. In 1994 and 1996, liquid chromatography mass spectrometry metabolomics experiments were performed by Gary Siuzdak while working with Richard Lerner (then president of the Scripps Research Institute) and Benjamin Cravatt, to analyze the cerebral spinal fluid from sleep deprived animals. One molecule of particular interest, oleamide, was observed and later shown to have sleep inducing properties. This work is one of the earliest such experiments combining liquid chromatography and mass spectrometry in metabolomics. In 2005, the first metabolomics tandem mass spectrometry database, METLIN, for characterizing human metabolites was developed in the Siuzdak laboratory at the Scripps Research Institute. METLIN has since grown and as of 2025, METLIN contains MS/MS experimental data on over 960,000 molecular standards and other chemical entities, each compound having experimental tandem mass spectrometry data generated from molecular standards at multiple collision energies and in positive and negative ionization modes. METLIN is the largest repository of tandem mass spectrometry data of its kind. The dedicated academic journal Metabolomics first appeared in 2005, founded by its current editor-in-chief Roy Goodacre.

Nevertheless, with onset of COVID-19, he continued to support, through ad hoc advice, on various assessments in regards to vaccine distribution. At NCCD, Kohli had key involvement in rationalizing and revising the cold chain support mechanism and various other related schemes to promote and encourage cold chain development. He also was prime mover in establishing a historic MoU of cooperation in cold chain between nodal Indian and French organisations. He was the French Government's guest of honor and represented India at the International Conference on Sustainable Cold Chain in Paris (April 2013). In July 2015 he was invited as a global expert to speak on the Global Food Crisis at the House of Lords in UK. He was regularly invited to lecture at the national academy for Indian administration services and the National Defence College from 2017 to 2022. Kohli was a member of various national committees and councilor to industry organisations and is chairman of the national Committee on Supply chain and Logistics, the first such committee set up with a focus on the supply chain by the government. He also co-chaired committees on Human Resource Development and Research & Development for cold chain. He was the principal member of the Indian government's Committee for Doubling Farmers' Income, set up in 2016, where his innovative percepts provoked a paradigm shift in how agricultural supply chain systems are understood. He has authored a number of policy documents and a key contributor to many game changing policies in the cold chain domain.
In 2010, the US government began cracking down on pharmacists and doctors who were overprescribing opioid painkillers. An unintended consequence of this was that those addicted to prescription opiates turned to heroin, a significantly more potent but cheaper opioid, as a substitute. A 2017 survey in Utah of heroin users found about 80 percent started with prescription drugs. In 2010, the Controlled Substances Act was amended with the Secure and Responsible Drug Disposal Act, which allows pharmacies to accept controlled substances from households or long-term care facilities in their drug disposal programs or "take-back" programs. In 2011, the federal government released a white paper describing the administration's plan to deal with the crisis. Its concerns have been echoed by numerous medical and government advisory groups around the world. In July 2016, President Barack Obama signed into law the Comprehensive Addiction and Recovery Act, which expands opioid addiction treatment with buprenorphine and authorizes millions of dollars in funding for opioid research and treatment. In 2011, the Obama administration began to deal with the crisis, and in 2016, President Barack Obama authorized millions of dollars in funding for opioid research and treatment, followed by CDC director Thomas Frieden stating that "America is awash in opioids; urgent action is critical." Soon after, many state governors declared a "state of emergency" to combat the opioid epidemic in their own states, and undertook major efforts to stop it.

=== Sports === According to Bloodworth and McNamee sports and physical activities are a key contributor to the development of people's well-being. The influence of sports on well-being is conceptualized within a framework which includes impermanence, its hedonistic shallowness and its epistemological inadequacy. Researching the effect of sport on well-being is difficult as some societies are unable to access sports, a deficiency in studying this phenomenon.

Advisors are large larvae-like aliens which are virtually featureless, with no visible eyes, ears or limbs, although they do possess an eye-like mechanical device attached to the left side of their heads, and detachable mechanical arms. Their faces are covered by a respirator, which is able to lift to reveal a mouth-like orifice from which they can extend a long and flexible proboscis. With this, they can examine objects, or attack and kill other creatures. Advisors appear to be feeding upon their victim during their attacks. It is implied that Advisors are the Combine's ruling class, with Breen answering directly to them. Although Advisors are usually seen in protective pods guarded by Combine soldiers, they also possess psychokinesis with which they are able to float through the air and immobilize other creatures so that their proboscis can examine victims without interference. Their appearance was based on the Guild Navigators from the 1984 film Dune.
===== Methane ===== Methane produced by marine methanogens is typically more D-enriched than methane produced by methanogens grown in freshwater. δDs for thermogenic methane range from −275‰ to −100‰, and from −400‰ to −150‰ for microbial methane.

The sequence is Met-Glu-His-Phe-Pro-Gly-Pro, a seven-residue peptide. The first four residues correspond to the ACTH(4-10) fragment, and Pro-Gly-Pro is added at the C-terminus. N-acetylated versions also appear in the literature and differ in mass.
Semax is a synthetic analog of the ACTH(4-10) fragment rather than a copy of the full hormone. Truncation plus the added tail removes the steroidogenic portion of ACTH. That distinction explains why it is studied for nervous-system effects instead of adrenal stimulation.
It is registered as a prescription product in Russia and has been used there clinically, chiefly by the intranasal route. No approval exists in the United States or the European Union. Availability elsewhere typically falls under research-chemical distribution, with varying purity documentation.
It is a synthetic seven-amino-acid peptide built from the ACTH(4-7) sequence plus a Pro-Gly-Pro tail. It belongs to the family of short ACTH fragments studied for nervous-system effects. Its backbone contains only standard amino acids.
