This is a working overview of lyophilization, written for readers who want more than a one-paragraph summary but less than a textbook.
This page was last updated on 2026-02-22 and is reviewed periodically as new material appears.
Verification of a supplied batch generally combines a certificate of analysis with independent testing, because certificates are self-reported documents. A typical package includes a chromatographic trace, a mass spectrum, and a stated water or counter-ion content. Batch-to-batch consistency matters more than a single purity figure when results are compared across experiments. No single mandatory standard governs research-grade peptide release, so laboratories are expected to define their own acceptance criteria. Residual trifluoroacetate from purification is a frequently overlooked counter-ion.
Lyophilized material is chemically stable for extended periods when kept dry, cold, and protected from light. The powder is hygroscopic, so vials should be warmed to room temperature before opening to reduce condensation on the contents. Once dissolved, the peptide is far less stable because peptide bonds are susceptible to hydrolysis and the methionine residue can oxidize. Solutions are typically aliquoted and held at 2-8 °C for short intervals or frozen for longer ones, and repeated freeze-thaw cycles should be avoided.
Later generations of the molecule include an N-acetylated form and an amidated form, both marketed online alongside the parent peptide. These variants differ in terminal chemistry and stability, and they are frequently discussed in the same breath even though they have not been compared in controlled trials. Supply outside formal healthcare systems comes largely from laboratories that synthesize peptides to order. Purity and identity of these materials vary widely, and no single body oversees the international trade.
Semax is a synthetic heptapeptide developed in the Soviet Union during the 1980s by researchers at the Institute of Molecular Genetics in Moscow. It was designed as a truncated analog of adrenocorticotropic hormone, retaining only the fragment spanning residues four through ten. Investigators sought a peptide that would preserve the cognitive effects associated with ACTH while eliminating the hormonal stimulation of the adrenal cortex. The compound entered clinical use in Russia during the following decade.

Claims about enhanced focus, memory, or mood in healthy people rest mostly on anecdotal reports and community discussion rather than on controlled data. It remains unclear whether any cognitive benefit observed in patients recovering from brain injury would extend to uninjured users. Dose-response relationships, long-term safety, and interactions with other drugs are not well characterized in the peer-reviewed literature. Questions about optimal route of administration and treatment duration likewise remain open.
Semax binds to melanocortin receptors and is thought to influence neuronal survival and plasticity rather than to act through the adrenal axis. Laboratory work has shown increased expression of brain-derived neurotrophic factor and nerve growth factor in treated tissue. Changes in c-Fos, a marker of neuronal activation, have also been reported. Because the peptide is rapidly degraded by peptidases, its effects are generally attributed to downstream signaling cascades rather than to sustained receptor occupancy.
Published studies are dominated by animal models of stroke, ischemia, and cognitive impairment, with a smaller number of human trials conducted in Russia. Many of the human reports are small, single-center, and published in Russian-language journals, which limits independent scrutiny. Outcome measures vary between studies and often rely on clinician-rated scales rather than objective biomarkers. Systematic reviews have noted the methodological weaknesses and called for larger, preregistered trials before firm conclusions can be drawn.
The C-terminal Pro-Gly-Pro extension is not incidental. Proline-rich tails are known to resist several common peptidases, and the published literature attributes the longer half-life of Semax, relative to unmodified ACTH fragments, to this feature. The modification also removes the melanocyte-stimulating and corticosteroidogenic activity that characterizes longer ACTH-derived sequences. Because the molecule is small and hydrophilic, it is typically formulated as an aqueous solution for intranasal or parenteral delivery. Acetylation or amidation at the termini appears in closely related research peptides and shifts the mass by a fixed increment.
Reported pharmacological work centers on neurotrophic signaling, including changes in BDNF and NGF expression in hippocampal tissue in animal models. Human data come largely from studies conducted in Russia, and how well those results generalize to other populations remains an open question. Regulatory status differs sharply by jurisdiction: Semax is a registered prescription medicine in Russia, while it holds no approved marketing status in the United States or the European Union. Outside such jurisdictions it is generally handled as a research chemical, which affects both documentation and quality expectations.
Semax is a synthetic heptapeptide whose sequence is Met-Glu-His-Phe-Pro-Gly-Pro. It was developed as a fragment analog of adrenocorticotropic hormone, modeled specifically on the ACTH(4-10) region. The first four residues reproduce that fragment, while a Pro-Gly-Pro tripeptide is appended at the C-terminus. Work on the compound originated in Russia, where it entered clinical use as an intranasal preparation. Its sequence places it among short regulatory peptides studied for effects on the central nervous system rather than on the adrenal axis.
Semax is normally supplied as a lyophilized powder, and that form is the most stable. Suppliers commonly recommend storage at -20 °C in a desiccated container protected from light, with short-term handling at room temperature limited to weighing and transfer. Powder that has absorbed moisture degrades faster, so vials should be warmed to ambient temperature before opening to prevent condensation on the contents. Hygroscopic behavior is typical of short hydrophilic peptides, and humidity control matters more than temperature alone for long-term retention.
Once dissolved, the peptide is considerably less stable than the solid. Aqueous solutions are usually prepared at neutral to slightly acidic pH, filtered, and divided into single-use aliquots before freezing. Repeated freeze-thaw cycles are a common cause of avoidable loss and are best prevented by never refreezing a thawed aliquot. Adsorption to plastic and glass surfaces can lower the measured concentration of dilute solutions, particularly below roughly 0.1 mg/mL. Buffer choice, salt content, and container material all influence how much peptide remains detectable after storage.
The compound was developed during the 1980s at the Institute of Molecular Genetics in Moscow as part of research on fragments of adrenocorticotropic hormone. Early work examined short ACTH-derived sequences that retained neurotrophic effects while lacking the endocrine activity of the full hormone. Semax entered clinical use in Russia during the 1990s, where it received registration for several neurological indications. Outside that region it remained primarily a laboratory research material rather than an approved therapeutic. English-language literature on it grew more slowly and frequently cited the original Russian studies.
Terminology around the compound varies by source. It appears in catalogues and papers as Semax, as the heptapeptide ACTH(4-7)-Pro-Gly-Pro, and under various alphanumeric laboratory codes used by individual suppliers. These names refer to the same sequence but may imply different salt forms, purity grades, or counter-ions. Peptide databases usually list the free base mass, while product descriptions sometimes report acetate or trifluoroacetate salts with a different formula weight. Because naming conventions for research peptides are not standardised across vendors, checking the declared sequence and measured mass is more reliable than relying on a trade name alone.
Semax is a synthetic heptapeptide with the sequence Met-Glu-His-Phe-Pro-Gly-Pro. Its design combines the ACTH(4-7) core fragment with a C-terminal Pro-Gly-Pro extension, a modification intended to improve stability and prolong activity. The molecule is hydrophilic, carries no lipid chains or glycosylation, and has a theoretical mass just over 810 daltons in its free form. All seven residues are proteinogenic amino acids, so no non-natural building blocks appear in the backbone. A free N-terminal methionine and C-terminal proline define the unmodified parent peptide.

This thus favors a shift towards Th2 dominance and the release of the cytokines listed above which results in Th2 dominance and favors the 'humoral' B-cell mediated antibody immune response. Cortisol also has a negative-feedback effect on IL-1. The way this negative feedback works is that an immune stressor causes peripheral immune cells to release IL-1 and other cytokines such as IL-6 and TNF-alpha. These cytokines stimulate the hypothalamus, causing it to release corticotropin-releasing hormone (CRH). CRH in turn stimulates the production of adrenocorticotropic hormone (ACTH) among other things in the adrenal gland, which (among other things) increases production of cortisol. Cortisol then closes the loop as it inhibits TNF-alpha production in immune cells and makes them less responsive to IL-1. Through this system, as long as an immune stressor is small, the response will be regulated to the correct level. In general, the hypothalamus uses cortisol to reduce the response once the production of cortisol matches the stress induced on the immune system.
=== Secondary dentin === Secondary dentin is formed after root formation is complete, normally after the tooth has erupted and is functional. It grows much more slowly than primary dentin but maintains its incremental aspect of growth. It has a similar structure to primary dentin, although its deposition is not always even around the pulp chamber. It appears greater in amounts on the roof and floor of the coronal pulp chamber, where it protects the pulp from exposure in older teeth. The secondary dentin formed is not in response to any external stimuli, and it appears very much similar to the primary dentin. It is the growth of this dentin that causes a decrease in the size of the pulp chamber with age. This is clinically known as pulp recession; cavity preparation in young patients, therefore, carries a greater risk of exposing the pulp. If this occurs, the pulp can be treated by different therapies such as direct pulp capping. Previously it was thought that pulp capping was most successful if followed by a stainless steel crown, however this procedure is usually unnecessary in children. It requires the unnecessary removal of enamel which is key to the life of the tooth. Adhesive dentistry allows for conservative restoration techniques that minimize the loss of tooth structure and should be used. In order to maintain space in the primary dentition, attempts are made not to extract a pulpal exposure.
== Iron-57 == 57Fe is widely used in Mössbauer spectroscopy and the related nuclear resonance vibrational spectroscopy due to the low natural variation in energy of the 14.4 keV nuclear transition. The transition was famously used to make the first definitive measurement of gravitational redshift, in the 1960 Pound–Rebka experiment.
=== Pregnancy and lactation === The use of dalbavancin in pregnant women has not been studied sufficiently and should only occur when the potential benefit outweighs the potential risk to the fetus. Animal studies did not show embryo or fetal toxicity at doses that were 1.2 and 0.7 times the human dose. However, delayed fetal maturation was observed at a dose that was 3.5 times the human dose. While dalbavancin is excreted in rat milk, it is unknown if it is excreted in human milk. It should be used in nursing mothers only when the potential benefit exceeds the potential risk. There is no evidence in animals of teratogenicity.

=== Sickle Cell === Sickle-cell disease changes the shape of red blood cells from round to sickle shaped. In the most common variant of sickle-cell disease, the 20th nucleotide of the gene for the beta chain of hemoglobin is altered from the codon GAG to GTG. Thus, the 6th amino acid, glutamic acid, is substituted by valine—notated as an "E6V" or a "Glu6Val" mutation—which causes the protein to be sufficiently altered with a sickle-cell phenotype. The affected cells cause issues in the bloodstream as they can become sticky due to their improper ion transport leading to them being susceptible to water loss. This can cause a buildup of blood cells that obstructs blood flow to any organ in the body.
== Digico Micro 16 == Digico quickly started developing a general purpose 16-bit minicomputer, the Micro 16, which became available in 1966. Digico was assisted by the Ministry of Technology and the National Research Development Corporation in this development. The first version produced was the Digico Micro 16S (1968), followed by the 16P (1970), then the 16V in 1972.

== Genetics == Chinese landraces were found to have a slightly higher genetic diversity than inbred lines by Li et al., 2010. Specific locus amplified fragment sequencing (SLAF-seq) has been used by Han et al., 2015 to study the genetic history of the domestication process, perform genome-wide association studies (GWAS) of agronomically relevant traits, and produce high-density linkage maps. An SNP array was developed by Song et al., 2013 and has been used for research and breeding; the same team applied their array in Song et al., 2015 against the USDA Soybean Germplasm Collection and obtained mapping data that are expected to yield association mapping data for such traits. Rpp1-R1 is a resistance gene against soybean rust. Rpp1-R1 is an R gene (NB-LRR) providing resistance against the rust pathogen Phakopsora pachyrhizi. Its synthesis product includes a ULP1 protease. Qijian et al., 2017 provides the SoySNP50K gene array.

=== Other === Labial fusion, also called labial adhesion, is the fusion of the labia minora. This affects a number of young girls and is not considered unduly problematic. The condition can usually be treated using creams, or it may right itself with the release of hormones at the onset of puberty. Clitoromegaly is an enlarged clitoris caused by either anabolic steroids or an intersex condition. Vulvodynia is chronic pain in the vulvar region. There is no single identifiable cause. A subtype of this is vulvar vestibulitis but since this is not thought to be an inflammatory condition it is more usually referred to as vestibulodynia. Vulvar vestibulitis usually affects pre-menopausal women. Pudendal nerve entrapment can cause sharp pain or numbness in the vulva. This condition can be caused by activities such as cycling, giving birth, or prolonged sitting. A number of skin disorders such as lichen sclerosus, and lichen simplex chronicus can affect the vulva. Crohn's disease of the vulva is an uncommon form of metastatic Crohn's disease, which manifests as a skin condition showing as hypertrophic lesions or vulvar abscesses. Papillary hidradenomas are nodules that can ulcerate and are mostly found on the skin of the labia or of the interlabial folds. Another more complex ulcerative condition is hidradenitis suppurativa, which is characterised by painful cysts that can ulcerate, and recur, and can become chronic lasting for many years. Chronic cases can develop into squamous cell carcinomas.
=== Part I: Democratic Rights === Explicitly drawing from the United Nations' Universal Declaration of Human Rights, Article One of the Charter declares that every individual is entitled to the following:"a. The freedom of conscience, expression, association, and peaceable assembly; b. The right to engage in unrestricted political activity and to organize political parties, provided the exercise of such right does not infringe upon the rights of others."Article Two concerns the rights of "nations, nationalities, and peoples" in Ethiopia, referring to the various ethnolinguistic groups in the country. Not only does the Charter protect nationalities' right to exercise their autonomy, but it also allows them to secede ("self-determination of independence") if they so wish.
=== Gas cloud composition === Apart from MIC, based on laboratory simulation conditions, the gas cloud most likely also contained chloroform, dichloromethane, hydrogen chloride, methylamine, dimethylamine, trimethylamine, and carbon dioxide that was either present in the tank or was produced in the storage tank when MIC, chloroform, and water reacted. The gas cloud, composed mainly of materials denser than air, stayed close to the ground and spread in a southeast direction affecting the nearby communities. The chemical reactions may have produced a liquid or solid aerosol. Laboratory investigations by CSIR and UCC scientists failed to demonstrate the presence of hydrogen cyanide.
The second proposed mechanism via an anhydride has similar steps but there is a direct attack of Glu270 on the carbonyl group, and then the interaction of Glu270 on the Zn2+-bound amide forms an anhydride instead which can subsequently be hydrolyzed by water.

Dry powder is normally held at -20 °C or lower, away from light and moisture. Sealed vials can also be kept at 2-8 °C for shorter intervals. Warming to room temperature before opening prevents condensation.
In solution, hydrolysis of peptide bonds and oxidation of the methionine residue are the principal concerns. Oxidation converts methionine to its sulfoxide form, shifting the mass by sixteen daltons. Both processes are slowed by cold storage and by avoiding alkaline conditions.
A certificate documents what the supplier measured on a specific lot, not how a laboratory will use the material. Independent verification of identity and purity is common practice. Acceptance criteria should be defined before a batch is ordered.
It originated at the Institute of Molecular Genetics in Moscow during the 1980s. The work was carried out by a Russian research group that specialized in peptide neuropharmacology.
