==== Ionic selectivity and plasticity ==== Several groups reinvestigated the permeability properties of TPCs and their role in NAADP-induced Ca2+ release, and they agreed that TPCs are indeed permeable to Na+ but they could not necessarily recapitulate the Na+ selectivity shown in the 2012/13 studies. It was therefore initially proposed that TPCs may conduct both Ca2+ and Na+ (analogous to the NMDA receptor of the plasma membrane). As more studies were published, why some groups observe a Na+ selectivity while others see a mixed Na+/Ca2+ permeability was unclear until the important realization that the TPC2 ionic selectivity wholly depended on the activating ligand. Currents activated by PI(3,5)P2 were predominantly carried by Na+ whereas NAADP-activated currents showed an eight-fold increase in the Ca2+ permeability. This conveniently explained the discrepancies between groups as well as revealing that TPC2 is extraordinarily plastic in operating in different conductance modalities. Since then, it appears that TPC2 can be synergistically activated by co-application of NAADP and PI(3,5)P2, although the molecular mechanisms are unclear. Therefore, TPC2 can operate as either a Ca2+ or Na+ channel, depending on whether NAADP or lipid activate it.

==== Multiple sclerosis-related fatigue ==== Fatigue is a common and often debilitating symptom experienced by people with multiple sclerosis. Reviews and meta-analyses of controlled trials have found that modafinil has modest effectiveness in managing MS-related fatigue, though improvements in fatigue severity scores have not consistently reached statistical significance. Optimal dosing and treatment schedules are not well established. Clinical assessments have found that adverse events were common. Most MS organizations are neutral on the off-label use of modafinil to alleviate fatigue associated with MS.
=== Engineered peptidyl transferase center === In 2014, it was shown that by altering the peptidyl transferase center of the 23S rRNA, ribosomes could be created which draw on orthogonal pools of tRNA. The 3' end of tRNAs is universally conserved to be CCA. The two cytidines base pair with two guanines the 23S rRNA to bind the tRNA to the ribosome. This interaction is required for translational fidelity. However, by co-mutating the binding nucleotides in such a way, that they can still base pair, the translational fidelity can be conserved. The 3'-end of the tRNA is mutated from CCA to CGA, while two cytidine nucleotides in the ribosomes A- and P-sites are mutated to guanidine. This leads to ribosomes which do not accept naturally occurring tRNAs as substrates and to tRNAs, which cannot be used as substrate by natural ribosomes. To use such tRNAs effectively, they would have to be aminoacylated by specific, orthogonal aaRSs. Most naturally occurring aaRSs recognize the 3'-end of their corresponding tRNA. aaRSs for these 3'-mutated tRNAs are not available yet. Thus far, this system has only been shown to work in an in-vitro translation setting where the aminoacylation of the orthogonal tRNA was achieved using so called "flexizymes". The legal status of AOD-9604 at the time of the program was uncertain: it was not explicitly listed as banned under Category S2 of the World Anti Doping Agency (WADA) code, but it had not been approved for human use by the Therapeutic Goods Administration and therefore could be considered as banned under the more general Category S0, which applied to any substances not approved for human use. The official position of WADA was that AOD-9604 had been considered a banned substance under Category S0 since 2011, and it clarified this publicly to ASADA on 22 April 2013. After considering its legal position, ASADA declared that it would not pursue any cases relating to AOD-9604 use before 22 April 2013, determining that it would be too difficult to achieve a guilty verdict for a supplement that had not yet explicitly been declared banned. === Legal status === In June 2009, a US Food and Drug Administration (FDA) advisory panel voted by a narrow margin to advise the FDA to remove Vicodin and another opioid, Percocet, from the market because of "a high likelihood of overdose from prescription narcotics and acetaminophen products". The panel also cited concerns of liver damage from their paracetamol component, which is also the main ingredient in commonly used nonprescription drugs such as Tylenol. Each year, paracetamol overdose is linked to about 400 deaths and 42,000 hospitalizations. In January 2011, the FDA asked manufacturers of prescription combination products that contain paracetamol to limit the amount of paracetamol to no more than 325 mg in each tablet or capsule within three years. The FDA also required manufacturers on all paracetamol-containing products to issue a black box warning indicating the potential risk for severe liver injury and a warning highlighting potential for allergic reactions. In August 2014, the Drug Enforcement Administration (DEA) announced that all hydrocodone combination products (HCPs) would be rescheduled from schedule III to schedule II of the Controlled Substances Act (CSA), effective in October 2014. == Properties == Despite the name, aerogels are solid, rigid, and dry materials that do not resemble a gel in their physical properties: the name is because they are made from gels. Pressing softly on an aerogel typically does not leave even a minor mark; pressing more firmly will leave a permanent depression. Pressing extremely firmly will cause a breakdown in the sparse structure causing it to shatter like glass (a property known as friability), although more modern variations do not suffer from this. Even though it is prone to shattering, it is very strong structurally. Its impressive load-bearing abilities are due to the dendritic microstructure in which spherical particles of average size 2–5 nm are fused together into clusters. These clusters form a three-dimensional highly porous structure of almost fractal chains, with pores just under 100 nm. The average size and density of the pores can be controlled during the manufacturing process. An aerogel material can range from 50% to 99.98% air by volume, but in practice most aerogels exhibit somewhere between 90 and 99.8% porosity. Aerogels have a porous solid network that contains air pockets, with the air pockets taking up the majority of space within the material. Aerogels are good thermal insulators because they almost nullify two of the three methods of heat transfer – conduction (they are mostly composed of insulating gas) and convection (the microstructure prevents net gas movement). They are good conductive insulators because they are composed almost entirely of gases, which are very poor heat conductors.
